K+-Cl?-cotransport Mediates the Deoxyribose 1-phosphate-Induced Melanogenesis

Abstract

|
The effect of deoxyribose 1-phosphate (dRP) on melanogenesis was investigated in B16 melanoma cells. dRPincreased both reactive oxygen species (ROS) levels and melanin content in a dose-dependent manner in B16 cells. Theseeffects were significantly reduced by apocynin (Apo) and diphenyleneiodonium (DPI), which are inhibitors of the NADPHoxidase (NOX). DIOA, a blocker of the K+-Cl?-cotransports (KCCs), significantly inhibited dRP-induced activation ofKCCs and melanogenesis. The NOX inhibitors(Apo and DPI) also significantly blocked dRP-induced activation of KCCs.
Collectively, these results suggest that dRP may activate KCCs through NOX-mediated ROS production, and in turn,induce melanogenesis in B16 cells.
